How to get into Bioinformatics?

Hi
I've been doing a lot of search on the internet on how to get started
in Bioinformatics, I was inspired by Drew Endy with his talk on 24C3:

and I was wondering what do I need to know to start? I have some
programming knowledge and know a little biology from high school. Am
currently doing a physics degree so that doesn't really help on bio.
But do I need to know a lot of Biochemistry inorder to do some DNA
sequencing?
Also what on earth is a DNA syntheizer? what you make your own DNA and
put it into a bacterial or something?
Any helpful comments appreciated
